NGS Libraries on Element Biosciences AVITI: Performance Compared with Illumina
The range of NGS instrument options keeps expanding beyond Illumina, and among them, Element Biosciences’ AVITI has drawn attention as a platform built around its own distinct chemistry and throughput. AVITI is designed so that its proprietary CloudBreak Freestyle technology can accept Illumina-compatible libraries directly for sequencing.
Evaluating Cross-Platform Performance
What Celemics set out to confirm in this study wasn’t simply whether sequencing was possible, but whether libraries built with Celemics’ library preparation kits and capture panels would deliver a comparable level of data quality on AVITI to that observed on Illumina. Even when cross-platform compatibility is in place, differences in practical performance metrics such as mapping rate, on-target ratio, or coverage uniformity can affect real-world use.
To test this, libraries built from two of Celemics’ custom capture panels, Panel A and Panel B, were loaded onto both an Illumina NextSeq 500 and an Element Biosciences AVITI system, sequenced, and compared using an identical analysis pipeline.
Comparable Sequencing Performance on AVITI and Illumina
The results showed no meaningful differences between the two platforms across mapping rate, duplication rate, on-target ratio, Fold-80 base penalty, and target coverage depth for both panels. These results demonstrate that libraries prepared with Celemics’ kits maintained data quality on AVITI comparable to that observed on Illumina.

One Library Prep Workflow, More Sequencing Options
These findings confirm that Celemics’ library preparation kits and capture panels demonstrated comparable performance on AVITI and Illumina in this evaluation. Customers looking to broaden their sequencing platform options without altering their established prep workflow can consider AVITI as an additional sequencing platform while maintaining the same library preparation workflow. This also provides a practical operational benefit. Labs running both Illumina and AVITI in parallel do not need to stock separate kits for each platform – a single kit inventory can support both sequencers, easing inventory management.
There is also no need to learn a separate library preparation protocol or re-optimize the preparation workflow; the same prep procedure can be applied to an AVITI run. Detailed metrics and data are available in the Celemics Application Note.
