MGI Library Preparation Method: Conversion from Illumina to MGI

Streamlined NGS Workflow: Convert Illumina Libraries for MGI Sequencing

Celemics provides an option that allows customers using both Illumina and MGI sequencing platforms to utilize a single Library Preparation (LP) and Target Enrichment (TE) kit for both platforms. This is made possible through our Illumina to MGI conversion method.
In this post, we will introduce the conversion process that enables sequencing of an Illumina-prepared library on the MGI platform, along with a performance comparison of the sequencing results.

Protocol: Celemics Library Preparation & Target Enrichment Procedure

The figure below illustrates the workflow for Celemics’ Native Library Preparation and Target Enrichment kits for both Illumina and MGI platforms.
As shown, the conversion method involves selecting different primers during the post-PCR step following the TE process. Depending on the primers used, the final library can be sequenced either on the Illumina or MGI platform.

Results

The experiment was conducted using three different samples per panel. After completing the TE process, we obtained on-bead captured libraries. Normally, only half of this is used for the final post-PCR. In this study, one half was amplified using Illumina primers and the other half using MGI conversion primers. Sequencing was then performed on the respective platforms. The results confirmed that converting a Celemics Illumina library for use on the MGI platform does not compromise performance. Therefore, by simply adding the MGI conversion option to the Celemics Illumina Library Preparation and Target Enrichment kits, users can conveniently switch between the two sequencing platforms.
