From RNA to Reliable Data: Why RNA Requires Careful Handling

To unlock the secrets of biological phenomena, we analyze RNA within cells to obtain information on gene expression. RNA is a crucial indicator of gene activity at a specific point in time, but it is structurally unstable and prone to degradation. For this reason, instead of handling RNA directly, many molecular biology experiments convert it into a more stable form—complementary DNA (cDNA)—through a process called reverse transcription. Particularly in downstream applications like Next-Generation Sequencing (NGS), the use of double-stranded cDNA (ds-cDNA) is standard practice. 1. Consistency Across Variables: From Different Users to Input Amounts

In fact, we have confirmed that consistent results are obtained even when the protocol is performed by experimenters with different skill levels. Furthermore, the kit shows robust performance across a wide range of input RNA amounts. Internal validation has shown that successful library preparation with similar patterns was achieved with input RNA amounts ranging from as low as 10 ng to as high as 250 ng. Subsequent Differentially Expressed Gene (DEG) analysis also yielded stable data without significant bias.

2. Performance: FFPE and Viral Samples

The kit’s applicability to actual clinical specimens has also been validated. For FFPE samples, a prime example of degraded RNA, experiments using reference standards not only resulted in the production of high-quality libraries but also led to the successful detection of all known fusion variants within the standard. Additionally, the kit was confirmed to perform effectively in experiments with actual viral specimens, demonstrating a high detection rate.
