Celemics Blocker: Improve your NGS Target Enrichment efficiency

In order to maximize the accuracy and efficiency of NGS, the Blocker plays a crucial role. Celemics designs and manufactures the Blockers, whether it is sequencer-specific or species-specific, used in all our Target Enrichment Kits to ensure our customers to achieve optimal NGS results. We would like to dedicate this post to introduce Celemics Blockers.

The Necessity of Blockers

 

When using Celemics’ Target Enrichment Kit, we guide our customers to include the Blocker during the Hybridization process for optimal performance. The efficiency and differences in sequencing result with and without the Blocker can be seen in the table below.

*This experiment used the Celemics Enzymatic Preparation Kit (EP Kit) for Illumina, preparing a library with 100 ng of HAPMAP Sample. Target Enrichment experiments were conducted using 500 ng of each library, and sequencing data analysis was normalized approximately to 2,000,000 reads. On-target ratio and other metrics may vary depending on the panel used.

As seen in the table, there is a significant difference of over 50% in the on-target ratio. Data with Blocker showed an average depth of 62.33 for the target region, while data without Blocker showed an average depth of 23.85 (data not shown), and the overall sequencing metrics clearly confirms that Blockers are needed for adequate performance.

Applications of Celemics Blockers

During Celemics’ Blocker manufacturing process, several practices are conducted to enhance stability and efficiency, providing reliable Target Enrichment performance. Using Celemics’ core manufacture technology, we can provide Blockers that are specifically designed for various sequencing platforms, as well as for various different species. We offer Blockers for well-known sequencing platforms such as Illumina, MGI, and Thermo Fisher (Ion Torrent), along with various species, including humans, pigs, mice, and plants like rice. Additionally, we are continuously developing and updating our Blockers to expand and adapt to other sequencing platforms and target species.
