CELEMICS Case Report: Library Conversion from Illumina to Singular Genomics

CELEMICS provides NGS kits optimized for various platforms, ensuring seamless integration for customers using different sequencing systems. This post is to introduce how Celemics kits can be tailored for Singular Genomics’ NGS platform.

Workflow

First, let’s explore how NGS library targeted for Illumina platforms can be converted for use on Singular Genomics platform.

 

The conversion process follows the same workflow as our existing Illumina library prep and target enrichment kits. This means that customers already using Celemics products can seamlessly transition to the Singular Genomics NGS platform without modifying their experimental procedures.

Results

We evaluated the performance of our conversion strategy by comparing libraries processed through our conversion method with native Illumina-prepared libraries.

Experiment Condition: Hybridization with 500 ng library. Panel size over 1 Mbp, Post-PCR 14 Cycles

After performing Target Enrichment and subsequent post-PCR or conversion PCR, we observed relatively equivalent experimental results. Next, we conducted primary analysis using our in-house bioinformatics pipeline to compare sequencing performance.

*This experiment was conducted using CELEMICS’ Enzymatic Library Preparation kit, Target Enrichment kit, and a Celemics Custom Panel. **The prepared libraries were sequenced on both the Illumina NextSeq and Singular Genomics G4 instruments. Analysis was performed using adapter removal, BWA, SAMtools, Picard, and GATK, with mapping to the GRCh37 reference genome.

Metrics such as Duplication Ratio, On-Target Ratio, Fold 80 Base Penalty, and Percentage of Uncovered Regions showed no significant differences between the Illumina and Singular platforms. These results confirm that Celemics’ Singular Genomics Kit follows the same library prep and target enrichment workflow as our Illumina kits, delivering high-quality sequencing results equivalent to those obtained on the Illumina platform.

Additionally, we offer library conversion kits for MGI, Element Biosciences, and PacBio Onso platforms.
