AgBio Research with NGS: Cost-Effective and Accurate Target Sequencing

Next-generation sequencing (NGS) technology allows for the rapid and efficient analysis of large volumes of nucleotide sequences, a method widely used across various fields of biology. In the agricultural biotechnology (AgBio) sector, NGS is utilized for purposes such as plant and animal breeding, genotype-based trait research, pathogen analysis, and microbiome studies. AgBio research often involves processing large numbers of samples, making cost-effectiveness a key consideration. In this regard, targeted sequencing, which focuses on specific genes or genomic regions, is gaining attention as an effective and economical tool in AgBio. In target capture kits, blockers play a crucial role in enhancing the accuracy and efficiency of NGS. If blockers are not used or their performance is inadequate, the on-target percentage of sequencing data decreases, meaning that more sequencing data must be generated to achieve the same level of analytical results, and ergo, the cost of analysis increases. To ensure optimal analytical performance, Celemics directly manufactures sequencing platform-specific and species-specific blockers used in target capture kits.

Molecular breeding is the process of selectively improving desirable traits and developing new varieties based on genetic information. Existing analysis methods such as GBS (Genotyping by Sequencing), Microarray, and PCR each have their advantages, but they also face limitations when analyzing large sample sizes or complex genetic information. For example, while GBS and Microarray are suitable for analyzing a large number of markers, GBS can have low coverage due to its dependence on restriction enzyme sites, and Microarray is limited to analyzing predefined markers. PCR, on the other hand, is effective for quickly analyzing a relatively small number of markers and is advantageous for processing large sample volumes but is less suitable for analyzing a large number of markers

Target sequencing offers flexible control over the number of markers, provides high coverage for the target regions, and supports high-throughput sample processing using automated equipment. This makes it possible to overcome the limitations of existing technologies and perform more precise and comprehensive analyses. Based on Celemics’ reagent production capabilities and custom kit manufacturing technology, we can create panels for molecular breeding markers of animals and plants as per customer requirements, and supply them along with optimized reagent sets.

In addition to panels for plant and animal genomes, Celemics also offers a variety of pathogen analysis panels. Pathogens that cause diseases in animal and plant species or zoonotic pathogens can be effectively detected through targeted sequencing. Depending on the characteristics of the pathogen and the testing environment, we can determine the analysis target from specific markers to whole genomes, and we can provide both Hybridization and PCR Amplicon-based panels. Based on our experience in developing various target capture panels, we have successfully performed sequencing on regions that were difficult to capture with conventional analysis methods.
